It just breaks down faster/easier if you add the amino acids to the water where as if you add water it tends to get stuck at the bottom and have to shake it more/harder. But no difference besides the sticking to bottom.

I would say add 1/8 of the powder, then 1/8 of the water, then another 1/8 of the water followed by a quarter of the powder, then 1/2 the water, the rest of the powder and very slooooowly add the rest of the water. Now if you want to know how to mix it properly while doing this, I'll need another couple of posts.
