Excuse if this is an amateurish question.
When they are referencing the amount of protein to eat to gain or lose weight how am I supposed to calculate. Am I to calculate based on the weight I want to be? Or my current weight? I.E. being 300 lbs and wanting to get to 215, or 215 getting to 230 lbs, etc.

It's a debated topic.
It's what works best for you so you maintain strength while on a deficit. I started cutting at 215, currently i'm down to 195ish. I consume about 1800 cal/day and about 160g of protein.
When I set my goals I went off my Lean Body Mass Estimate. So far my body fat is down and my strength is high.----------------------------------------------
6'2 - Begin @ 215lb. - 12/23/11 @ 197lb. - 1/19/12 @ 194lb.
Goal - 185lb @ 10-12% bf.
