I've had a lot of experience with weight training kids from my high school and training a few kids at college. They've ranged from short to tall and fat to skinny, but they've all been guys. When I get back home from college I plan to train my sister and cousin, but I'm at a bit of a loss at how to go about this.
My sister in particular wants to work on:
1. Flexibility
and
2. Getting a "nice round ass"
She doesn't want to get too "jacked", but I don't think that's too much of an issue because girls aren't oozing out testosterone. I'll probably stretch the crap out of them before moving onto a strength based split, but is there anything else I should know about before I start?

What would your training plan have been if you had one of the kids at college who wanted to increase their flexibility and get a 'nice round ass'???
What would your training plan have been if a male had approached you wanting to increase their flexibility and get a 'nice round ass'???
Would you be at a loss at how to train either of these examples? What are the major difference between all three scenarios?

So you know how to work out. Great. You can be a workout partner for your sister and cousin. But please don't claim to call yourself a "trainer".
The simple fact you are going to stretch them before they start working with weights means you should probably not be getting paid for your advice. You're probably better off printing out the "booty shaping" routine from Women's Health and doing it with them.

Don't do any developmental stretching pre-strength and conditioning.
Structure a suitable programme for their specific goals - as someone else said, they're not aliens, they're human beings who will respond to training in their individual way and will need individual goals to attain their individual results.
Warm-up
Pre stretch appropriate muscles (don't force stretch - only hold for 10 seconds)
Muscle strength and toning (squatting, lunging etc) 10-15 rep range.
Cardio
Cool down
Developmental stretching (hold until tension eases, extend stretch slightly and hold for 30s) repeat.
If you're not a trainer then I guess you won't be doing any kind of pre-assesment for medications or injuries etc. Get them to see a doctor first I realise that they are family and you might know this, but better be safe than sorry.Go Bravely, without fear, in the direction of your dreams.
