A knot is a muscle spasm, I think you are confused about what you have. If you do truly have a spasm in your TA, which is very rare, then use heat an massage to treat it. This will losen up and relax the muscle.
Shin splints is pretty much a garbage term for a lot of conditions. Shin splints could be a strain of the TA, a stress fracture of the tibia, the muscle pulling off of the bone, etc. Generally treatment included icing and rest.
If you have just started running on the treadmill, it's going to take some time to get used to it. Changing running surface is the usual culprit of "shin splints"
