MICROBIOLOGY ASSAY!! I need HELP (Substrate COncenrtration vs OD)
Sample work problem.
Substrate concentration (mM)
( Tubes were made with the following concentrations of SUbstrate...(any substrate)
In mM 10 20 30 40 50 60 70 80 90
time0 0.005 0.005 0.004 0.005 0.005 0.00 0.004 0.005 0.295
48hrs 0.095 0.17 0.23 0.26 0.275 0.28 0.295 0.295 0.296
QUESTION TO PROBLEM IS:
You have a sample of some food that when this bacterium grows on the extract it reaches an OD of 0.205 after 48hrs and shows not further increase. When started it had an OD of 0.005. How many moles of substrate did the extract contain?
I have managed to do a graph that looks like this
OD Reading l
(yaxis) l
l
l an upside down parabola goes here!!!
l
l
l________________
(x-axis) [S] (Substrate concentration( the difference from time 0 to after 48hrs of incubation)
I added and subtracted the difference in substrate concentration, from beginning to ending
These are the results I got
from the 10mM all the way through 90mM
=
In mM for tube with 10 mM.009
In mM for tube with 20 mM.165
In mM for tube with 30 mM = .226
In mM for tube with 40mM =.255
In mM for tube with 50 mM =.270
In mM for tube with 60 mM = .28
In mM for tube with 70 mM =.291
In mM for tube with 80 mM = .29
In mM for tube with 90 mM =.001
I plotted these on the x axis, against the OD reading (Data given at the top, for each tube)
I have no idea how to answer the question it is asking...!
What should I do,how do I solve this problem??????????
PLEASE Help Me!!!

It is the absorbance, or reading of a machine ( Machine is called and spectrophotometer)
It is used to read how much light was deflected or absorbed by the machine, this reading can tell you how much or an organism you have in a sample, which is inside a cuvette, a special squared tube

Well you can determine the molarity of a solution by its absorbance on a spectrophotometer no? I don't really know how to do it I didn't do Biol4s I'm only at Biol200 lol and MIMM212 and 211 ...
but try to look at this website
http://www.chm.davidson.edu/ronutt/c...ro/Spectro.htm
sorry. good luck!
