I recently tried a harder shoulder workout by adding more sets and finishing with partials.
Felt great the day of, but the next day it felt like I had DOMS in my rotator cuff muscles.
Is it possible for those muscles to have DOMS?

Doesn't sound right to me but I suppose it is possible. It could also be a strain.

It's absolutely possible to have DOMS in rotator cuff muscles. Any muscle is susceptible to it.
Are you certain, though, that it is a soreness which replicates DOMS? or, a soreness which may lean more towards injury?
I ask this, because, the pain of a muscle strain is a completely different feeling to DOMS.

yea, And I get DOMS fairly often. It usually comes the next day and goes away after 2 days. This is the same case. My shoulder already feels better.
And I find if its a muscle strain it will hit you much sooner than DOMS.
Yeah, in general you will feel a strain or overworked muscle nearly instantly depending on the degree of the strain.
DOMs in the RC muscles is like DOMS in all other muscles... usually intensifying 24-48 hours later and lasting past 72 hrs sometimes.
Don't overwork your shoulders. RC muscles are pretty small so they can easily get injured. If they're getting sore a lot you may need to back off the volume some.
just fyi, DOMS can be generated pretty much anywhere,

Generally speaking, DOMS is brought on by unfamiliar action or load in an *eccentric* contraction.
